چکیده
20 We demonstrate a simple method for combining instant structured illumination microscopy (SIM) 21 with total internal reflection fluorescence microscopy (TIRF), doubling the spatial resolution of TIRF 22 (down to 115 +/13 nm) and enabling imaging frame rates up to 100 Hz over hundreds of time points. 23 We apply instant TIRF-SIM to multiple live samples, achieving rapid, high contrast super-resolution 24 imaging in close proximity to the coverslip surface. 25 26 . SNSMIL, a real-time single molecule identification and localization algorithm for super-resolution fluorescence microscopy
Single molecule localization based super-resolution fluorescence microscopy offers significantly higher spatial resolution than predicted by Abbe's resolution limit for far field optical microscopy. Such super-resolution images are reconstructed from wide-field or total internal reflection single molecule fluorescence recordings. Discrimination between emission of single fluorescent molecules a...
متن کامل* Single - molecule fluorescence imaging by total internal reflection fluorescence microscopy ( IUPAC Technical Report )
Total internal reflection fluorescence (TIRF) is a popular illumination technique in microscopy, with many applications in cell and molecular biology and biophysics. The chief advantage of the technique is the high contrast that can be achieved by restricting fluorescent excitation to a thin layer. We summarise the optical theory needed to understand the technique and various aspects required f...
متن کاملA Thin Layer Imaging with the Total Internal Reflection Fluorescence Microscopy
Total internal reflection fluorescence microscopy (TIRFM) is an optical technique that allows imaging of a thin layer of the sample with a thickness of about 100-200 nm. It is used in science of cell biology to study cellular processes, especially near the membranes of living cells. This method is based on the total internal reflection phenomenon, where the evanescent wave is generated in the l...
متن کاملTracking sub-microscopic protein organisation at the plasma membrane of live cells using triple-colour super- resolution microscopy
Cell signalling is regulated by submicroscopic features of the plasma membrane, including the actin cytoskeleton and clathrin-coated pits. Total internal reflection microscopy is ideal for studying this, but resolution is limited to approximately 200 nm. This article explains how, at the University of Osnabrück, we have established a triple-colour super-resolution imaging approach to attain an ...
متن کاملSuper-Resolution Imaging of Mitochondria and Microtubules
Super-resolution microscopy using the TIRFM (total internal reflection fluorescence microscope) was employed to visualize and elucidate the interactions between mitochondria and microtubules inside the cell. The mitochondria were labeled with the enhanced yellow fluorescent protein (EYFP) through transfection, and the microtubules were labeled with the synthetic dye, Alexa Fluor 647, through im...
